2) I am planning to hook the two 1/4" John Guest fittings to doser pumps. Is a check valve required to prevent return-side pressure on these fittings (i.e., I am worried about pressure from the return pump being directed through these fittings towards the doser pumps)?

Yes I would dry fit the plumbing first then glue all fitting under the aquarium. I would not suggest using the reactor feed lines to connect dosing pumps as those lines will be pressurized. Personally I would exclude that whole manifold setup completely unless you plan to use a calcium reactor or small media filter as it was intended for.
